Folding, Reasoning, and Scaling with Open-source Drug Discovery Engine

Aureka AI OpenDDE project

Accurately modeling biomolecular interactions is a central bottleneck in biology and therapeutic discovery. Here, we introduce Open Drug Discovery Engine (OpenDDE), an open-source, all-atom biomolecular foundation model that uses co-folding as the entry point to a scalable AI-driven drug discovery engine. Rather than treating structure prediction as an isolated endpoint, OpenDDE is designed as a shared structural reasoning layer for modeling sequence-structure-function relationships across biomolecular complexes, enabling complex structure prediction today while providing a foundation for de novo design, affinity estimation, structure-conditioned optimization, and more. OpenDDE integrates advances in all-atom architecture, atomic latent reasoning, inference optimization, and large-scale data processing to achieve IsoDDE-level co-folding accuracy within a reproducible and openly accessible framework. We also identify two scaling-law directions for co-folding models, revealing practical routes for continued improvement through data, model, inference, and training scaling. By releasing training code, inference pipelines, checkpoints, and benchmarks, OpenDDE aims to democratize access to frontier biomolecular intelligence, accelerate global collaboration, and lay an open foundation for next-generation drug discovery systems that can move from predicting molecular structures toward designing, scoring, and optimizing therapeutic candidates for human health.

Unsupervised Consensus-Based Anomaly Detection for Spatiotemporal Malaria Incidence in Ghana

T. Ansah-Narh, Y. Asare Afrane

A consensus anomaly detection framework was applied to monthly malaria surveillance data from Ghana (2014-2023) to identify atypical transmission patterns. Anomalies were highly structured in space and time. Ashanti and Northern Regions accounted for most recurrent anomalies, wit…

MEUSLI: a Multilingual Projector for LLM-based ASR and Beyond

Lorenzo Concina, Seraphina Fong, Marco Matassoni, Alessio Brutti

Lightweight projectors are an established way to connect pre-trained speech encoders with large language models (LLMs), mapping acoustic features into token-level embeddings for tasks like ASR and spoken question answering. Existing systems, however, typically only support a few…

Agentic Method for Deterministic Validation of Legacy Code Migration

Andras Ferenczi, Jordan Docherty, Mariya Bessonov, Matthew Findlay, Krishna Lingamneni

Migration of legacy COBOL programs to Java requires extensive testing to ensure correct functionality. This effort is often complicated by the lack of test data and the difficulty of validating all corner cases. In this paper we propose a novel agentic test-synthesis method, the…

Hardware-Software Co-Design for Float16 On-Device Training on RISC-V Single-Core

Benjamin Hubinet, Pierre-Alain Moellic, Olivier Savry, Olivier Potin, Jean-Baptiste Rigaud

By leveraging standard RISC-V extensions, namely Zfh (scalar float16) and Zvfh (vector float16), this work proposes an open-source framework to enable complete on-device training on resource-constrained RISC-V single-core. Our approach allows memory footprint reduction by about 5…

Multi-primitive in-memory computing for Monte Carlo tree search

Tergel Molom-Ochir, Benjamin F. Morris, Yintao He, Archit Gajjar, Giacomo Pedretti, Hai Helen Li, et al.

Monte Carlo tree search (MCTS) enables artificial intelligence (AI) decision-making, but requires 55-300 W on conventional processors, limiting edge deployment. In-memory computing (IMC) is energy-efficient on regular workloads but has been considered incompatible with irregular…
